Development of the Multiplex PCR for Detection of the DNA-Contained Emergent Diseases Agents in Pigs (African Swine Fever, Aujeszky Disease, Circoviral Disease)

چکیده
Introduction African swine fever virus (ASFV), pseudorabies virus (Aujrszky’s disease virus, ADV) and porcine circovirus type 2 (PCV-2) are some of the most dangerous DNA viruses causing high amounts of morbidity and mortality in commercial and backyard pig farms. Traditional diagnosis of porcine viral infections requires complex and lengthy combinations of virological and serological tests. This study aimed to develop a method for rapid detection of the DNA-containing viral pathogens of pig in clinical materials using conventional multiplex PCR platform.

Detection Of Toxoplasma Gondii and Human Cytomegalovirus DNA in Blood from Transplant Recipients Using Multiplex Nested Polymerase Chain Reaction
Evidences from many studies suggested a polymerase chain reaction (PCR) as a valuable method for diagnosing infectious disease in the transplant recipients. We used this method for detection of Toxoplasma, gondii and human cytomegalovirus in blood specimens from patients after bone marrow or kidney transplantation. DNA of both infectious agents were detected using two separate sets of nested pr...
متن کاملA highly sensitive and specific gel-based multiplex RT-PCR assay for the simultaneous and differential diagnosis of African swine fever and Classical swine fever in clinical samples.
The development and standardisation of a novel, highly sensitive and specific one-step hot start multiplex RT-PCR assay is presented for the simultaneous and differential diagnosis of African swine fever (ASF) and Classical swine fever (CSF). The method uses two primer sets, each one specific for the corresponding virus, amplifying DNA fragments different in length, allowing a gel-based differe...
متن کاملDesign and verification of a highly reliable Linear-After-The-Exponential PCR (LATE-PCR) assay for the detection of African swine fever virus.
African swine fever virus (ASFV) is a highly pathogenic DNA virus that is the causative agent of African swine fever (ASF), an infectious disease of domestic and wild pigs of all breeds and ages, causing a range of syndromes. Acute disease is characterized by high fever, haemorrhages in the reticuloendothelial system, and a high mortality rate. A powerful novel diagnostic assay based on the Lin...
متن کاملDevelopment of a Multiplex RT-PCR Assay for Detection of the Causal Agents of Citrus Tristeza and Cachexia Diseases with Coamplification of Plant mRNA as an Internal Control
Background and Aims: Plant certification programs need reliable, fast, cheap and sensitive methods for detection of systemic pathogens with special interest in virus and viroid detection. Reverse transcriptase-polymerase chain reaction (RT-PCR) has been documented as an alternative assay for certification of plant propagating materials. The main object of the present study was the optimization ...
متن کاملDevelopment and optimization of Multiplex- PCR for simultaneous detection of Porcine Pseudorabies Virus, Porcine Parvovirus, and Porcine Circovirus Type 2
Multiple infections by pathogens are currently the most serious problems in pig herds. Clinically, accurate diagnosis is difficult due to similarity of the symptoms of porcine pseudorabies virus (PRV), porcine parvovirus (PPV), and porcine circovirus type 2 (PCV2). A multiplex polymerase chain reaction (multiplex-PCR) was developed and optimized for the simultaneous detection of the three DNA v...
